Sample of B.frutescents leaf were collected throughout Batu Rakit in Kuala Terengganu. Leaves of B.frutescens extracted with methanol solvent. The total weight of B. frutencens leaves crude extract obtained was 2.532 g from 10 g of initial weight were subjected to chemical compound analysis solvent such as Methanol, Acetonotrile and water through qualitative High Performance Liquid Chromatography (HPLC) profiling analysis and were screened for antibacterial and cytotoxicity properties. The profile spectrums from HPLC showed a few peaks that represented the chemical that lie in the methanolic extract especially in methanol. The result from HPLC crude extract profile for leaves extract B.frutescens showed many peaks at the retention time between 0 to 60 minutes. Two bioactivity of studied were antibacterial and cytotixicity assay. In antibacterial assay, six species pathogenic bacteria, that are, Escherichia coli, Morganella morganii, Serratia marcescens, Salmonella typhii, Listeria monocytogenes, and Klebsiella pneumonia were selected and used by using agar well diffusion method. All methanolic extract did not show any antibacterial activity or was not effective at all concentration of 12.5-100mg/ul. As a result, methanolic extract was not subjected to broth dilution method for the quantitative measurement of the microbiostatic (inhibitory). However, six species of pathogenic bacteria have shown a tremendous growth for all different concentrations. For cytotoxic assay, the cell lines that were used were HL-60, MCF-7 and HT-29 that were treated with B.frutescens methanolic extract to determine the IC50 value, a concentration of test compounds that causes 50% inhibition or cell death. It is aimed to comparethe cytotoxic effects between them with 3T3 which acted as a negative control. Result showed that B.frutescens methanolic extract was not effective for normal cell line. The methanolic extract of B. frutescens was cytotoxic to HL-60, MCF-7, and HT-29. Their IC50 of the crude extract were moderate between 17.6 ug/mL respectively. Extract of B.frutescens was most cytotoxic against HL-60 with IC50 value 17.6 ug/mL.
