description Amylase is one of the most important enzymes in industrial application. The study investigated the production and characterization of produced by NSH9 isolates under solid state fermentation. The study showed cassava was the best substrate for amylase inducer. The highest amylase production detected at 72 hours of incubation with enzyme activity is 4.3170 IU/ml and the protein concentration was 0.0104 mg/ml. The best pH for amylase production was pH7.0 with 4.3170 IU/ml and 4.3170 IU/ml for 27°C of incubation temperature. The protein concentration for pH and temperature was 0.0104 mg/ml and 0.0104 mg/ml. The enzyme was analyzed by using dinitrosalicyclic acid (DNS) assay method. The characteristic of enzyme activity was showed the highest activity at pH6 (1.7325 IU/ml) and at 37°C (1.6998 IU/ml) after 10 minutes incubated with 1% (w/v) soluble starch. The kinetic study of the amylase produced was shown the value of Vmax is 0.4743 mg ml-1min-1 and Km is 679.86 mg ml-1.
