| description | The Diamond-Back Moth,(Plutella xylostella (Linnaeus) Lepidoptera: Yponomeutidae) is a major pest of crucifers (Brassicaceae) crops throughout the world (Reddy & Urs, 1996). Many researches have been done about their resistance to pesticides. Most of them are focused on the attempt to control this species of pest for example the studies on the sex pheromone of the diamondback moth Pilitella xylostella (Lepidoptera: Yponomeutidae) in India by Reddy & Urs (1996). DNA was extracted using C-TAB protocol and amplified using PCR targeting on Cytochrome P450. The P450 monooxygenases has several functions that include physiological, honnone and pheromone metabolism (Berembaum, Cohen and Schuler, 1990). These monooxygenases are a diverse group of enzymes specifically encoded from a superfamily comprising at least 17 families, and probably many more (Nebert & Gonzales, 1987). Modifications were done on homogenisation method and centrifugation time. Overall the C-TAB protocol was suitable to extract DNA from samples after modification on the homogenization method and centrifugation time with an overall percentage of success rate 43.75%. All PCR amplifications of the samples did not yield results. |
