description A comparative analysis are made to compare flowsheet designs of the application of Aqueous Two-Phase System (ATPS) to extract protein from cell lysate. The analysis that observes the protein purity and yield trends is conducted using a flowsheet calculation framework that calculates the thermodynamic behaviour of the polymers and polymer-salt systems containing protein(s) within a limited range of properties and concentration. The designs include one-stage and two-stage extraction stages containing two types of ATPS i.e. PEG6000-DxT500 and PEG6000-Na3PO4 containing phosphofructokinase and ovalbumin. The results match previous findings to some extent. The analysis highlights potential for flowsheet design optimization work.
