description Abstract: Objectives: This study was aimed to examine the effect of two different media on bone marrow stem cells (BMSCs) chondrogenesis in vitro and in vivo. Methods: BMSCs were isolated from sheep and divided into two test groups and control group. Control group was cultured in medium supplemented with 10 % fetal bovine serum (FBS). Test group A was cultured in medium supplemented with 1% FBS, 1% ITS, 5ng/mL IGF-1, 3ng/mL bFGF and 1ng/mL TGF-β2. Test group B was cultured in medium supplemented with 1%FBS, 1% ITS, 5ng/mL TGF-β3, 50ng/mL IGF-1, 40 ng/mL L-proline, 100 nM dexamethasone and 50 μg/mL ascorbic acid-2 phosphate. The experiment was carried out in Institute of Medical Research (IMR) and Universiti Kebangsaan Medical Centre Malaysia, Kuala Lumpur city, from 19 April 2007 to 25 December 2008. Results: Cell aggregates were formed in test group A and more in test group B denoted the early chondrogenesis in culture. Later, cells-fibrin constructs were made and implanted subcutaneously into nude mice for 5 weeks, then explanted for histological examination and glycosaminoglycans (GAG) quantification. Test group B constructs showed highest cartilage matrix synthesis confirmed by Safranin O staining and GAG production. Conclusion: These results demonstrated the effectiveness of medium supplemented with TGF-β3 in the combination to promote chondrogenesis in BMSCs.
