However, SOXD and SOXE functions and mechanisms in brain tumors in East Coast of Peninsular Malaysia is still lacking. This study was aimed to determine the brain tumor distribution in East Coast of Peninsular Malaysia and to evaluate the SOXD and SOXE gene expression of brain tumor in this region. This study then predicted the roles of SOXD and SOXE protein in biological processes, molecular functions, cellular components, protein-protein interaction and pathways related to brain tumor by bioinformatic tools. A total of 217 brain tumor cases in Pathology Department of Hospital Universiti Sains Malaysia (HUSM) were selected from written reports dated 2011 to 2014. The distribution according to histological types and selected sociodemographic variables were evaluated by using Microsoft Excel and IBM SPSS version 22. RNA extraction was performed to 15 formalin-fixed, paraffin-embedded (FFPE) specimens of selected brain tumors including two non-neoplastic brain tissues as controls. SOX5, SOX6 and SOX13 from SOXD subgroup and SOX8, SOX9 and SOX10 from SOXE subgroup gene expression were generated by using qRT-PCR and analysed by using comparative Ct method. Finally, bioinformatics analysis was performed to obtain information on functions, mechanisms and pathways shared by SOXE and SOXD especially in neuronal development. Meningeal tumors were found to be the most common brain tumor with 36.4 % in this study followed by diffuse astrocytic and oligodendroglial tumors (18.4 %) and secondary brain tumour (12.9%). Gender preponderance was not significant in most of brain tumors types except meningioma whereby females were seen dominating the cases. Furthermore, increasing trend of brain tumor cases was observed across increasing age groups from 14.4% to 49.8%. Gene expression analysis of SOXD and SOXE in meningioma, glioma and secondary brain tumors revealed the expression status in this region. SOX6 and SOX13 (SOXD) was shown to be downregulated in most of brain tumor types. In contrast, SOX9 (SOXE) displayed upregulation in all of the brain tumor types except glioma. Gene Ontology (GO) analysis described SOXD and SOXE protein involvement in neurogenesis and it was supported by STRINGS that showed significant in interaction among SOXD and SOXE protein which reflected close interaction between these two gene subgroups. Pathway analysis by REACTOME showed the involvement of SOX6, SOX13 and SOX9 proteins in WNT/β-catenin pathway by tumor suppression or initiation capacity. This study has provided brain tumor status in East Coast of Peninsular Malaysia. SOX6 and SOX13 (SOXD) and SOX9 (SOXE) were found to play important roles in brain tumorigenesis via co-activation or/and co-repression and this result was supported by Gene Ontology analysis and pathway analysis.
