Eurycoma longifoliaor “tongkat ali” has long been claimed to improve strength of male virility and sexual prowess. Nowadays, numerous products from the plant have been introduced into the market as performance drink and herbal medicine to increase energy level as well as sexuality. However, scientific evidence to prove that this plant enhances sexuality in male and energy level is still lacking. In E. longifolia, normally only the root part was used for therapeutic purposes but stems and leaves were discarded. This practice makes the supply of E. longifolia raw material to be unsustainable and producing huge wastes of prime materials. Therefore, the objective of this study was to assess on aphrodisiac and ergogenic effects in stem and leaf extracts. The raw materials were first analyzed for physicochemical test. A total of six type of extracts were prepared from stem and leaf using different methods; leaf water extract (maceration), leaf water extract (reflux), leaf ethanol (50%) extract (Soxhlet), stem water extract (maceration), stem water extract (reflux) and stem ethanol (50%) extract (soxhlet). The extracts were screened and quantified with four selected markers: 13α (21)-Epoxyeurycomanone (EE), Eurycomanone (EU), 14, 15β dihydroxyklaineanone (DK) and Scopoletin (SP) using High Performance Thin Layer Chromatography (HPTLC) and High Performance Liquid Chromatography (HPLC). A new HPLC method for evaluation of markers content was developed and validated. The contents of primary and secondary metabolites in the extracts were determined by calorimetric methods and HPLC for amino acid. In vivo study of aphrodisiac effect was tested by penile erection using conscious male white rabbits. Ergogenic effect was established by forced swimming test in male mice. HPLC analysis showed the presence of EU and DK in all stem and leaf extracts. The extracts showed high content of glycosaponins, polysaccharides, proteins and presence of flavonoids, phenolics and phospholipids.Amino acid analysis showed stem maceration extract has high content of L-arginine, L-methionine and glycine. The results reveals significant effect, p < 0.05 when compared with control and it showed that stem maceration extract dose of 300 mg/kg prolonged the erection as compared to leaf extract. While, higher dose of extract, (625 mg/kg) showed significant increase, p < 0.05 in swimming time in mice after 49 days administration of stem maceration extract. The finding in this study showed that stem extract has high potential as aphrodisiac and ergogenic aids. This study also provides new data on the stem and leaf part of E. longifolia as it is no or too little investigated before.
