This study investigates the influence of different growth media and watering regimes on the propagation, physiological response, and bioactive compound content of Mas Cotek plants. The research was conducted to optimize the cultivation conditions for maximizing the production of bioactive compounds in Mas Cotek, which has been used in traditional medicine for its various health benefits. Methodology: In the study of rooting media composition, firstly the effects of rice husk biochar (RHB), BRIS soil, cocopeat (CC), peatmoss, and a mixture of 7CC: 3RHB as rooting media for the propagation of Mas Cotek cuttings was investigated. Secondly, the optimal ratio of CC and RHB for Mas Cotek cutting propagation was determined. Thirdly, the cutting propagation performance of Mas Cotek cutting was evaluated when propagated in the mixture of 7CC: 3RHB enriched with different rooting-promoting materials consisting of effective microorganism (EM), AgromediaTM, Indole-3-butyric acid (IBA), organic fertilizer, peatmoss, and SeasolTM. The cutting performance measured was the total number of roots, root length, plant biomass, root biomass, chlorophyll content, and stem diameter. The rooting media characteristics recorded were media pH, media electrical conductivity, and moisture content. Later the study on plant performance and bioactive compound as affected by media composition and watering volumes on the growth of Mas Cotek using the hanging bag technique was conducted. In this study treatments consisting of five different compositions of cocopeat (CC) and rice husk biochar (RHB) ratios of 100%CC:0%RHB, 70%CC:30%RHB, 50%CC:50%RHB, 30%CC:70%RHB and 0%CC:100%RHB and five different watering volumes of 400mL, 800mL, 1200mL, 1600mL and 2000mL were factorially tested. Data recorded were the media's physical and chemical properties, the plant's vegetative growth and physiological characteristics, and the bioactive compound (vitexin and isovitexin) in the leaf extract. Results: After one month of propagation, RHB was the best outcome in terms of the growth of plant roots which were total roots (12.2) and root length (14.48cm), and root biomass (1.130g). From the second propagation, it also found that Mas Cotek had the most total roots (11.4) and the longest root length (14.72cm) when propagated in 100% RHB. From the third propagation study, it was found that Mas Cotek was propagated best in the combination of 7CC:3RHB rooting media added with IBA which total roots (13.0) and root length (16.22cm). After 12 months of planting, Mas Cotek grown at 100%RHB and watered with 1600mL showed the best plant performance compared to other treatments. Conclusion: The studies concluded that propagation of Mas Cotek using rooting media 7CC:3RHB with the addition of suitable rooting-promoting material enhances the cutting performance. Findings also indicated that suitable growing media and water volume could increase plant performance for commercial purposes.
