This plant traditionally used to treat ringworms throughout the world and found wildly in tropical countries. The purpose of this study was to examine the in vitro cytotoxicity and genotoxicity effect of aloe-emodin on human promyelocytic leukemia cells (HL-60). For cytotoxicity assessment, there were several experiments done such as MTT assay, double staining AO/PI, DNA fragmentation, proliferation and annexin-V FITC. Meanwhile for genetoxicity assessment, alkaline comet assay was done by analysed each single cells after treated with aloe-emodin. For early screening purpose, MTT assay was used to determine the cytotoxic dose of several compounds on cancerous cell lines and normal cell line. Among the cancer cells lines, aloe-emodin showed strong killing effects against HL-60 cells. In contrast, aloe-emodin did have a mild cytotoxic effect towards mouse fibroblast NIH/3T3 cells. Thus, the HL-60 cells only were used to determine the mode of cell death after treated with aloe-emodin. The mode of cell death was mainly apoptosis assessed by acridine orange/propidium iodide (AO/PI) double staining method and Annexin V FITC/PI assays. Apoptotic cells characteristics such as the presence of blebbing cells and forming of apoptotic bodies were observed after HL-60 cells treated with aloe-emodin and examined using light microscope and fluorescence microscope. The mode of cell death was determined by AO/PI staining followed by flow cytometry. It was found that aloe-emodin induced cell death via apoptosis in the concentration dependant manner. The aloe-emodin did induce the PS of HL-60 cells to translocate from inner membrane cells to outer membrane cells. These exposures of PS attract the Annexin V conjugated with FITC to bind with the PS. Activation of Annexin V-FITC was allowed the flow cytometer to recognize HL-60 cells as an apoptotic cells. The percentage of cells that undergo apoptosis by using flow cytometer was almost at same percentage of cells undergo apoptosis in AO/PI assay. The effect of aloe-emodin on apoptosis was further assessed by DNA fragmentation. The formation of ladder pattern was observed in the samples treated with aloe-emodin. To corroborate this result, comet assay was performed to analyse the DNA damage on every single HL-60 cells treated with aloe-emodin. A comet tail like cells was observed after electrophoresis process which indicate DNA damage phenomenon.
